I had a quick fiddle with it. I think it might be something to do with the size of piece that is traced. I was tracing a phrase. As I increased the despeckle the smaller pieces, like the middle of the E where ignored. The higher the setting the bigger the pieces that were ignored. Not easy to explain it but I think if you were tracing something with marks around...I.e. not a clean image this could be used to filter the bits out.
it takes out some of the "interference" you may see in some traces - if you have ever taken a photo with a low resolution camera at night you will notice the speckled effect you can get. I always increase the size of my work surface when I play around with despeckle as you can then see what is happening
